The Effect of Dimethylformamide on the Liver of Albino Rat


Abstract
The toxicity of dimethylformamide in experimental animals has been studied by several investigators, -and it has also been reported to cause abdminal pain and hepatic, abnormalities in a fabric coating worker by an accidental dermal and respiratory exposure to dimethylformamide clinically. However, the precise mechan ism by which dimethylformamide causes the hepatic damage has not been understood in morphological aspect.
The author has demonstrated the effect of dimethylformamide on the liver, ob-serving the histological changes of the liver of albino rat treated with dimethyIformamide.
The experimental animals were given 1.5m1 per kg of body weight of dimethylformamide diluted with physiological saline solutionby intraperitonealinjection. The each of the experimental animals was hilled. at the intervals of 6, 12 :and 24¢¥ hours, -respectively. The histological preparations were stained with hema-toxylin and eosin method for histological changes of the liver.
The dimethylformamide treated rats showed the cellular swelling accompanied by vesicular degeneration and cellular necrosis associated with pyknosis and kary orrhexis of the hepatic parenchymal cells in the periportal zone of the hepatic lobule.
Consequently it is suggested that dimethylformamide would induce the vesicular degenertion and cellular necrosis of the hepatic parenchymal cells.
